In the experimental setup, the collimated laser beam is focused with a low numerical aperture lens (NA=0.025) into the center of a glass cuvette of square cross section. The low numerical aperture lens guarantees a depth of focus of 1 mm. The depth of focus is comparable to the optical path inside the cuvette (1 mm). The beam waist is calculated to be approximately 32 μm in diameter. Consequently the is approximately 1 nl. Nanoparticles of 300 nm (Streptavidin coated Bio-AdemBeads, AdemTech) were measured with a blue laser (405 nm, Nichia NDV4212T, operating at 120 mW). Nanoparticles of 500 nm (Streptavidin coated Masterbeads, AdemTech) were measured with a red laser (658 nm, Sanyo DL-6147-240, operating at 40 mW).
